中文摘要: 为评价沙棘多糖(polysaccharide from Hippophae rhamnoides L.,PHR)对环磷酰胺(cyclophosphamide,CTX)导致免疫抑制小鼠的免疫功能低下的缓解作用,以分离制备的PHR 为供试品,将小鼠分为空白组、环磷酰胺模型组、PHR 低、中、高(100、200、400 mg/kg)剂量组。利用环磷酰胺建立免疫抑制小鼠模型后,灌胃PHR,利用体质量称量、碳廓清、酶联免疫吸附(enzyme-linked immunosorbent assay,ELISA)、甲基噻唑基四唑盐比色法[(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay,MTT)]测定各组动物免疫器官指数、吞噬指数、血清免疫球蛋白G(immunoglobulin G,IgG)、免疫球蛋白M(immunoglobulin M,IgM)、肿瘤坏死因子α(tumor necrosis factor-alpha,TNF-α)、γ干扰素(interferon-gamma,IFN-γ)、白细胞介素6(interleukin-6,IL-6)及脾淋巴细胞增殖能力。结果表明,与空白组比较,模型组免疫器官指数、吞噬指数、血清中IgG、IgM、TNF-α、IFN-γ、IL-6 含量、淋巴细胞增殖能力均极显著降低(P<0.01),与模型组比较,PHR 各剂量组对以上指标均有所改善,其中PHR 中、高剂量组的作用效果显著(P<0.05)。因此PHR 可以缓解环磷酰胺导致的免疫抑制小鼠的免疫功能低下。
Abstract:To evaluate the immunoregulatory effect of polysaccharide from Hippophae rhamnoides L. (PHR) on immune dysfunction in cyclophosphamide (CTX)-induced immunosuppressed mice,PHR isolated and prepared from H. rhamnoides L. was used as the test sample. Mice were divided into blank group,CTX model group,and low-,medium-,and high-dose PHR groups (100,200,and 400 mg/kg). The immunosuppressed mouse model was established using CTX,followed by PHR administration via gavage. The immune organ index,phagocytosis index,serum levels of immunoglobulin G(IgG),immunoglobulin M (IgM),tumor necrosis factor-alpha (TNF-α),interferon-gamma (IFN-γ),interleukin-6 (IL-6),and the proliferation ability of spleen lymphocytes in each group were measured using weighing,carbon clearance,enzyme-linked immunosorbent assay (ELISA),and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The results showed that compared with the blank group,the model group exhibited significantly reduced immune organ index,phagocytosis index,serum levels of IgG,IgM,TNF-α,IFN-γ,IL-6,and lymphocyte proliferation capacity(P<0.01). Compared with the model group,each dose group of PHR showed improvement in the above indexes,with the medium-and high-dose groups demonstrating significantly greater effects (P<0.05). Therefore,PHR can alleviate immune dysfunction in CTX-induced immunosuppressed mice.
