血红素肽铁的絮凝分离工艺优化及其抗氧化活性

中文摘要: 为实现血红素肽铁高效分离提取,探究絮凝剂对血红素肽铁制备的影响。该研究以猪血为原料,以血红素肽铁得率及纯度为指标,筛选合适的絮凝剂,采用单因素(絮凝剂添加量、絮凝pH 值、絮凝温度、离心温度和离心时间)结合响应面试验优化絮凝分离工艺,并对其抗氧化活性进行表征。结果表明,选用质量浓度为1.5%的壳聚糖溶液、添加量为20%、絮凝pH5.60、絮凝温度44 ℃、离心转速13 100 r/min、离心时间为20 min 时,血红素肽铁的得率和纯度较高,分别为68.07%和30.57%。在此工艺下,血红素肽铁展现较强的抗氧化活性,其DPPH 自由基清除能力的IC50 值为1.17 mg/mL,ABTS+自由基清除能力的IC50 值为1.25 mg/mL,铁螯合能力的IC50 值为1.16 mg/mL。
Abstract:This study was performed to efficiently separate and extract heme peptide iron and explore the effects of flocculants on the preparation of heme peptide iron. The study used pig blood as the raw material and screened various types of flocculants based on the yield and purity of heme peptide iron. Single-factor experiments (including flocculant dosage, flocculation pH, flocculation temperature, centrifugation temperature,and centrifugation time) combined with response surface experiments were conducted to obtain the optimal flocculation separation process. The antioxidant activity of the obtained product was characterized. The results showed that using a 1.5% concentration chitosan solution with a dosage of 20%, flocculation pH of 5.60, flocculation temperature of 44 ℃, centrifugation speed of 13 100 r/min, and centrifugation time of 20 min achieved the highest yield and purity of heme peptide iron, reaching 68.07% and 30.57%, respectively. Under this process, heme peptide iron exhibited strong antioxidant activity. Specifically, the IC50 value for DPPH radical scavenging, ABTS+ radical scavenging, and iron chelating abilities were 1.17, 1.25 mg/mL, and 1.16 mg/mL,respectively.
