乌梅多糖提取工艺、理化性质及其体外功能活性

中文摘要: 该文主要研究乌梅多糖的提取工艺及其体外功能活性,考察超声功率、提取时间、料液比、提取温度对乌梅多糖得率的影响,采用响应面优化出乌梅多糖的最优提取工艺为料液比1∶22(g/mL)、提取温度39 ℃、提取时间67 min、超声功率160 W,乌梅多糖的平均得率高达9.7%,与预测值相近,采用sevage 法脱蛋白,对乌梅多糖的化学组成分析,扫描电镜结果显示其为片层结构,红外光谱结果表明其为α-构型呲喃糖,紫外光谱显示乌梅多糖不含有核酸、蛋白质、色素等。刚果红试验表明乌梅多糖不含三股螺旋结构,抗氧化活性试验结果表明乌梅多糖具有良好的抗氧化能力,乌梅多糖浓度为5 mg/mL 时,对甘氨胆酸钠的结合率达到45.73%,对牛磺胆酸钠的结合率为32.50%;乌梅多糖浓度为10 mg/mL 时,胰脂肪酶抑制率为49.7%,说明乌梅多糖具有良好的降血脂能力。
Abstract:The extraction process and in vitro functions of the polysaccharide from Fructus mume were studied.The extraction conditions including ultrasonic power,extraction time,solid-to-liquid ratio,and extraction temperature were optimized by response surface methodology.The optimal extraction process was a solid-liquid ratio of 1∶22(g/mL),extraction temperature of 39 ℃,extraction time of 67 min,and ultrasonic power of 160 W.The average yield of polysaccharide extracted from Fructus mume under the optimum conditions was 9.7%,which was close to the predicted value.The results of scanning electron microscopy showed that the extracted polysaccharide appeared a lamellar structure,and the infrared spectrum showed that the polysaccharide was α-morphopyranose.UV results showed that there was no nucleic acid,protein or pigment in the polysaccharide.The results of Congo red test showed that the polysaccharide had no triple helix structure and exerted high antioxidant activity.The polysaccharide at 5 mg/mL showcased the binding rates of 45.73% and 32.50% to sodium glycocholate and sodium taurocholate,respectively. The polysaccharide at 10 mg/mL demonstrated the inhibition rate of 49.7% on pancreatic lipase.The results suggested that Fructus mume polysaccharide had a lipid-lowering activity.
