产角蛋白酶芽孢杆菌50-3 的分离与初步鉴定

中文摘要: 为获得产角蛋白酶活性较高的菌株,以1 942 株动物粪便源芽孢杆菌为对象,在角蛋白酶筛选培养基中进行初筛,以菌株生长的透明圈与菌落直径比值较大的菌株在种子培养基中进行复筛,并进行菌株鉴定。结果表明:复筛得到的菌株50-3 在鸡毛粉作为角蛋白酶诱导底物的培养基上,37 ℃发酵36 h,角蛋白酶活力可以达到(694±15)U/mL。通过形态观察、生理生化分析、全细胞蛋白电泳和16S rDNA 序列分析,初步鉴定菌株50-3 为枯草芽孢杆菌进化分支中的一个种,命名为Bacillus sp.50-3。

Abstract:A total of 1 942 strains of Bacillus from animal feces were cultured for the preliminary screening of the strains with high keratinase activity. The strains with large ratios of hydrolysis zone diameter to colony diameter were re-screened in seed media and identified. The results showed that strain 50-3 fermented at 37 ℃for 36 h in the medium with only chicken hair powder for inducing keratinase production reached the keratinase activity of(694±15)U/mL. According to morphological characteristics,physiological and biochemical properties,whole-cell protein electrophoresis,and 16S rDNA sequencing,strain 50-3 was preliminarily identified as a species in the clade of Bacillus subtilis in the phylogenetic tree and named Bacillus sp.50-3.
