高效液相色谱法测定3 种市售鸡蛋中褪黑素含量

中文摘要: 该试验建立高效液相色谱配紫外-可见光检测器检测鸡蛋中褪黑素含量的方法,并对市售乌鸡蛋、柴鸡蛋及普通鸡蛋的蛋清、蛋黄中褪黑素含量进行测定。色谱条件:C18 色谱柱(4.6 mm×250 mm,5 μm);流动相0.05%三氟乙酸(trifluoroacetic acid,TFA)水溶液∶甲醇=40∶60(体积比),流速0.6 mL/min,进样量10 μL,检测波长222 nm,柱温30 ℃。结果表明:褪黑素浓度在0.1~10 μg/mL 时线性关系良好,标准曲线方程为y=128.57x-3.158 8,相关系数R2=0.999 9。方法检测限为0.019 6 μg/mL,定量限为0.065 2 μg/mL。蛋黄中褪黑素平均回收率为97.59%~98.69%,相对标准偏差(relative standard deviation,RSD)为0.04%~0.12%;蛋清中褪黑素平均回收率为96.94%~99.28%,RSD 为0.07%~0.49%。使用该方法对3 种市售鸡蛋的蛋黄、蛋清分别进行检测,结果显示,乌鸡蛋、柴鸡蛋和普通鸡蛋蛋黄中的褪黑素含量分别为39.50、39.70 ng/g 和38.56 ng/g,乌鸡蛋和柴鸡蛋蛋黄中褪黑素含量极显著高于普通鲜鸡蛋(P<0.01),乌鸡蛋、柴鸡蛋和普通鸡蛋的蛋清中褪黑素含量分别为10.70、10.74 ng/g 和10.28 ng/g,乌鸡蛋和柴鸡蛋蛋清中褪黑素含量极显著高于普通鸡蛋(P<0.01)。该方法回收率较高,精密度高,特异性强,峰形良好,适用于测定鸡蛋中的褪黑素含量。
Abstract:This experiment used high-performance liquid chromatography with a UV visible detector to establish a detection method for melatonin content in eggs.Determination was conducted on the melatonin content in the egg whites and yolks of a certain brand of commercially available dark eggs,native eggs,and ordinary fresh eggs.Chromatographic conditions:C18 column(4.6 mm×250 mm,5 μm);the mobile phase:0.05% trifluoroacetic acid(TFA);aqueous solution∶methanol=40∶60(volume ratio),with the flow rate of 0.6 mL/min,the sample size of 10 μL,the detection wavelength of 222 nm,and the column temperature of 30 ℃. The results showed that melatonin had a good linear relationship in the concentration range of 0.1-10 μg/mL. The standard curve equation was y=128.57x-3.158 8,and the correlation coefficient was R2=0.999 9.The limits of detection and quantification were 0.019 6 μg/mL and 0.065 2 μg/mL,respectively. The average recovery rate of melatonin in egg yolk was 97.59%-98.69%,and the relative standard deviation(RSD)was 0.04%-0.12%.The average recovery rate of melatonin in egg white was 96.94%-99.28%,while the RSD was 0.07%-0.49%.This method was applied to detect the yolk and egg white of three kinds of commercially available eggs with the same brand. It was found that the melatonin content in fresh egg yolk of dark eggs,native eggs,and ordinary eggs was 39.50,39.70 ng/g,and 38.56 ng/g,respectively. The melatonin content in the egg yolk of dark eggs and native eggs was significantly higher than that of ordinary fresh eggs(P<0.01). The melatonin content in fresh egg white of dark eggs,native eggs,and ordinary eggs was 10.70,10.74 ng/g,and 10.28 ng/g,respectively. The content of melatonin in egg white of dark eggs and native eggs was significantly higher than that of ordinary fresh eggs(P<0.01). With a high recovery rate,high precision,strong specificity,and good peak shape,the method is suitable for determining melatonin content in eggs.
