中文摘要: 通过动物实验评价薏苡仁提取物对睡眠的改善作用,并初步探讨其可能机制。50 只6 周龄雄性C57BL/6小鼠随机分为空白组(蒸馏水)、褪黑素组[褪黑素0.5 mg/(kg·d)]、薏苡仁干预组3 组[83.3、166.7、333.3 mg/(kg·d)]。连续灌胃30 d,每10 d 进行延长戊巴比妥钠睡眠时间实验、戊巴比妥钠阈下剂量催眠实验、巴比妥钠睡眠潜伏期实验;干预结束后通过居家笼实时监测小鼠睡眠行为学改变;酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测小鼠血清5-羟色胺(5-hydroxytryptamine,5-HT)、γ-氨基丁酸(γ-aminobutyric acid,GABA)、白介素-1β(interleukin-1β,IL-1β)和血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量。结果表明,薏苡仁提取物对小鼠无直接睡眠作用;干预30 d 后,中、高剂量组小鼠睡眠时间极显著延长(P<0.01);30 d 时各干预组小鼠入睡数显著增加(P<0.05);干预各组睡眠潜伏期有缩短趋势;居家笼监测干预组小鼠睡眠时间明显延长且存在剂量-反应关系;干预组血清与空白组相比IL-1β、5-HT、GABA 和TNF-α 含量均有升高趋势;IL-1β 含量与5-HT 含量、TNF-α 含量呈正相关;TNF-α 含量与GABA 含量呈正相关;5-HT 含量与GABA 含量呈正相关。薏苡仁提取物可以延长睡眠时间,增加入睡数,改善作用无依赖性且有持续效果,可能通过升高IL-1β、5-HT 和GABA 含量改善睡眠质量。
Abstract:To evaluate the effect of coix seed extract in improving sleep in mice and discuss the possible mechanism of action,fifty 6-week-old male C57BL/6 mice were randomly divided into five groups,including a blank group(distilled water),melatonin group[melatonin of 0.5 mg/(kg·d)],and three coix seed intervention groups at doses of 83.3,166.7 mg/(kg·d),and 333.3 mg/(kg·d).The experiments of prolonged pentobarbital sodium sleep duration,pentobarbital sodium subthreshold dose hypnosis,and pentobarbital sodium sleep latency were performed every 10 days by continuous intragastric administration for 30 days. After the intervention,the sleep behavior of mice was monitored by the home cage in real time. Enzyme-linked immunosorbent assay(ELISA)detected serum levels of 5-hydroxytryptamine(5-HT),γ-aminobutyric acid(GABA),interleukin-1β(IL-1β),and tumor necrosis factor-α(TNF-α).The results showed that coix seed extract had no direct effect on sleep in mice. After 30 days of intervention,mice in the medium and high dose groups of coix seed slept longer(P<0.01). On the 30th day,the proportion of sleeping mice in the intervention group increased significantly(P<0.05). Sleep latency showed a trend of shortening in each intervention group. Home cage monitoring revealed a significant increase in sleep time of mice in the intervention group,exhibiting a dose-response relationship.The serum levels of IL-1β,5-HT,GABA,and TNF-α were higher in the intervention group. IL-1β was positively correlated with 5-HT and TNF-α. TNF-α was positively correlated with GABA.5-HT was positively correlated with GABA.Coix seed extract could prolong the sleep time of mice and increase the number of sleeping mice.The improvement effect was independent and sustainable.The study suggested that coix seed may enhance sleep quality by elevating IL-1β,5-HT,and GABA in the serum.
