中文摘要: 以物理方法破壁后的滇黄精粉、茯苓粉、薏苡仁粉为原料,采用湿法制粒压片,以复合含片的感官评分和崩解时限为综合评分指标,利用单因素及正交试验确定含片最佳配方。运用Box-Behnken 响应面法优化制备工艺条件,同时评价其活性成分含量及体外抗氧化能力。结果表明,原料中薏苡仁粉、茯苓粉、滇黄精粉的质量比3∶3∶1,原料与辅料的质量比1∶4,辅料中乳糖与麦芽糊精的质量比1∶1,润湿剂乙醇体积浓度70%,46 ℃干燥25 min,过16 目筛后加入1.1%硬脂酸镁压片,该条件下制备的含片综合评分最高,其多糖含量104 mg/g,黄酮含量3.17 mg/g,体外抗氧化试验显示10 mg/mL 含片水提物对DPPH 自由基、ABTS+自由基、羟自由基的清除率分别为23.2%、32.0%、45.0%,总还原力为0.191,0.4 mg/mL 含片水提物对Fe3+的还原能力较VC 偏弱。复合含片成型完好,口感适宜,含服方便,具备一定营养价值。
Abstract:In this paper,the tablet was prepared by wet granulation method with the powder of Polygonatum kingianum,Poria cocos,and Coix lacryma-jobi whose cell walls were physically broken as the raw material.Taking the sensory evaluation and disintegration time of the compound tablet as the comprehensive scoring index,this paper adopted the single factor and orthogonal test in determining the best formula for the buccal tablet.The Box-Behnken response surface methodology was used to optimize the preparation conditions and evaluate its content of active components and in vitro antioxidant activity.The results showed that the comprehensive score of the buccal tablets reached the highest when the mass ratio of Coix lacryma-jobi powder,Poria cocos powder and Polygonatum kingianum powder in raw materials was 3∶3∶1,the mass ratio of raw materials and auxiliary materials was 1∶4,the mass ratio of lactose and maltodextrin in auxiliary materials was 1∶1,the volume concentration of ethanol(moistening agent)was 70%,and 1.1% magnesium stearate was added after the materials were dried at 46 ℃for 25 min and sieved through 16 mesh. Under this condition,the polysaccharide content of the tablet was 104 mg/g,the flavonoid content was 3.17 mg/g. In vitro antioxidant tests showed that the scavenging rate of DPPH free radical,ABTS+ free radical,and hydroxyl free radical by 10 mg/mL water extract of the tablet was 23.2%,32.0%,and 45.0%,respectively,and the total reducing power was 0.191. Compared to vitamin C,0.4 mg/mL water extract of the tablet showed a weaker reducing power to Fe3+. In conclusion,the compound tablet with certain nutritional value not only had a well-formed shape and delicate taste but also was convenient for administration.
