中文摘要: 采用组织块法对准噶尔山楂果实中的内生真菌进行分离,采用形态学与分子生物学结合的方法对果实中的内生真菌进行鉴定。利用琼脂块和滤纸片法进行抑菌试验,筛选出具有抑菌活性的菌株。采用薄层层析法(thin layer chromatography,TLC)初步分析发酵液中的有效成分,确定抑菌有效成分后,利用高效液相色谱法(high performance liquid chromatography,HPLC)对其中的有效成分进行含量测定。结果表明:共分离获得内生真菌14株,其中SZ-C菌株及其发酵液均对金黄色葡萄球菌和大肠杆菌具有较强的抑菌活性,TLC结果显示SZ-C的发酵液中含有槲皮素和山奈酚,HPLC检测结果显示SZ-C的发酵液中含有槲皮素和山奈酚含量分别为1.253 mg/L和1.329 mg/L。根据形态学特征和分子生物学分析确定SZ-C为极细链格孢菌。说明准噶尔山楂果实中存在能产生槲皮素和山奈酚的内生真菌并且具有一定抑菌效果。
Abstract:Endophytic fungi were isolated from junggar hawthorn fruit using a tissue block method and identified by morphological and molecular biology methods.Agar block method and filter paper were used to screen fungal strains for antibacterial activity.Thin layer chromatography(TLC)was used for preliminary analyses of active components in fermentation broths.After identifying effective antibacterial fractions,the content of the active fractions was determined by high performance liquid chromatography (HPLC).Fourteen strains of endophytic fungi were isolated,included SZ-C strains which produced conditioned media with strong antibacterial activity against Staphylococcus aureus and Escherichia coli.TLC results showed that the SZ-C conditioned media contained quercetin and kaempferol.HPLC analyses indicated that quercetin and kaempferol content were 1.253 mg/L and 1.329 mg/L.According to morphological characteristics and molecular biological analysis,SZ-C was identified as a very small Streptococcus bacteria.The results also indicated the presence of endophytic fungi in junggar hawthorn fruit that possess certain antibacterial effects and could produce quercetin and kaempferol.
