中文摘要: 传统的肉制品动物源性成分检测对检测人员操作水平及试验条件等方面要求较高,不适于基层检测需要。多酶恒温快速扩增技术(multienzyme isothermal rapid amplification,MIRA)是一种新型的核酸扩增方式,整个反应在37℃条件下即可完成,相比昂贵的聚合酶链式反应(polymerase chain reaction,PCR)仪器,检测成本以及试验条件要求明显降低。该研究通过结合多酶恒温快速扩增技术与侧向流层析技术(lateral flow dipstick,LFD),建立一种鸭源性成分多酶恒温侧流式扩增技术(MIRA-LFD)的快速检测方法,并对其特异性、检出性以及应用的可行性进行评价。

Abstract:Traditional methods for detection of meat and meat products require relatively high levels of personnel operation and expensive experimental procedures;these requirements can′t be met in grass-roots testing.Multienzyme isothermal rapid amplification(MIRA)is a type of nucleic acid amplification whereby the entire reaction can be completed at 37 ℃.Compared with the polymerase chain reaction(PCR)instrument,detection and testing using MIRA are significantly less expensive.In addition,the detection criteria are considerably relaxed.In this study,a MIRA-lateral flow dipstick (LFD)method was developed for rapid and simple detection of duck-derived materials.Moreover,the specificity,detection limit and feasibility of on-site testing of duckderived materials were evaluated through experiments.
