黑曲霉B1401固态发酵茶叶加工废料产单宁酶的研究

中文摘要: 为促进茶叶加工废料的高效利用,对黑曲霉B1401利用茶叶加工废料固态发酵生产单宁酶进行初步研究。在单因素研究的基础上,运用Plackett-Burman试验和Box-Behnken响应面法优化黑曲霉B1401固态发酵产单宁酶的最佳工艺条件。结果表明:可溶性淀粉0.48%、氮源比(尿素∶硝酸钠)0.74∶0.26(g/g)、接种量30%、装载量5 g、液固比1.8∶1(mL/g)、发酵温度30℃、发酵时间为99 h,在此条件下单宁酶酶活力可达到46.06 U/g。验证试验值44.02 U/g与模型的预测值基本相符,表明该试验方法适合于黑曲霉B1401的固态发酵工艺。
Abstract:The production of tannase by Aspergillus niger B1401 via solid-state fermentation was investigated using tea processed waste as a substrate.The Plackett-Burman experiment and Box-Behnken response surface method were used to optimize tannase production.Tannase production reached 46.06 U/g using the optimized conditions of soluble starch 0.48%,urea∶nitrogen ratio of as the nitrogen source 0.74∶0.26 (g/g),inoculum 30%,loading amount 5 g,liquid:solid ratio 1.8∶1(mL/g),fermentation temperature 30℃,and fermentation time 99 h.Of note,this experimental result was close to the predicted value 44.02 U/g according to response surface analysis,indicating that the method was suitable for solid-state fermentation process by Aspergillus niger B1401.
