中文摘要: 采用脂多糖(lipopolysaccharide,LPS)/氨基半乳糖(D-galactosamine,D-GalN)诱导建立小鼠急性肝损伤模型,用不同剂量蓝莓叶总黄酮(flavonoids of blueberry leaf,FBL)进行干预后,采集小鼠肝脏和血液,制备肝脏HE病理切片;检测血清中天冬氨酸转氨酶(aspartate aminotransferase,AST)和丙氨酸转氨酶(alanine aminotransferase,ALT)活性,以评价FBL对小鼠肝功能的影响;采用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测血浆中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子α(TNF-α)水平,以评价FBL的抗炎活性;测定肝脏组织中丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)、超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)的活性,以评价FBL的抗氧化作用。结果显示,与模型小鼠相比,FBL可显著降低LPS/D-GalN诱导的AST和ALT水平增高,明显抑制LPS/D-GalN引起的炎症因子分泌,有效逆转LPS/D-GalN引起的氧化损伤,并且有效缓解LPS/D-GalN导致的肝脏改变。上述结果表明,FBL对LPS/D-GalN诱导的急性肝损伤有保护作用,保护作用与抗炎和抗氧化作用有关。

Abstract:Male mice were intraperitoneal injected with lipopolysaccharide(LPS)/D-galactosamine(D-GalN)to induce acute liver injury after flavonoids of blueberry leaf(FBL)administration.The livers and bloods were collected after LPS/D-GalN injection to measure antioxidative activity and cytokines.The activities of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in serum were detected to evaluate the functions of the liver.The levels of inflammatory cytokines tumor necrosis factor alpha(TNF-α),interleukin-1 beta(IL-1β),andinterleukin-6(IL-6)weremeasuredusingenzyme-linkedimmunosorbentassay(ELISA)toevaluate the anti-inflammatory activity of FBL.The activities of malondialdehyde(MDA),glutathione(GSH),superoxide dismutase(SOD),and catalase(CAT)in liver tissues were analyzed to evaluate the antioxidative effect of FBL.The activities of AST and ALT were dramatically decreased in the FBL groups,compared with the model mice.FBL significantly repressed the secretion of inflammatory cytokine as a result of LPS/D-GalN administration.FBL treatment efficiently reversed the oxidative insult caused by LPS/D-GalN.The liver alterations induced by LPS/D-GalNwere efficiently relievedby FBLgroup.The resultsindicatedthatFBLpossessed the hepatoprotective effects by inhibiting inflammation and oxidative stress on LPS/D-GalN-induced acute liver injury.
