中文摘要: 以橡子壳斗为原料,采用超声辅助乙醇溶液法进行提取,在单因素试验基础上采用响应面法探究最优工艺条件,并测定pH 值、金属离子对橡子壳斗提取物溶液稳定性的影响,DPPH 自由基清除率及总还原力。结果表明影响提取物产率的因素为提取温度、提取溶剂浓度、料液比。利用Box-Behnken 试验法,确定最优工艺条件为提取温度60.0 ℃、溶剂浓度40%、料液比1 ∶35.5(g/mL),验证试验与预测值相吻合,且最优工艺下提取率为26.3%。橡子壳斗提取物在酸性环境中颜色变浅,碱性环境中颜色加深,呈棕红色。K+、Na+、Ca2+对提取物溶液无影响;Mg2+、Zn2+影响较小;Fe3+、Cu2+、A13+影响较大。提取物对DPPH 自由基的清除率略低于VC;铁离子总还原力略高于VC。
Abstract:Acorn shell was extracted by ultrasonic-assisted ethanol solution,and the optimum technological conditions were studied by response surface methodology (RSM)on the basis of single factor experiment.The results showed that the factors affecting extraction yield were extraction temperature,extraction solvent concentration and ratio of material to liquid.The optimum extraction conditions were determined by Box-Behnken test.The optimum extraction temperature was 60.0 ℃,solvent concentration was 40%,and the ratio of material to liquid was 1 ∶35.5(g/mL),the optimal extraction rate was 26.3%.Acorn shell extract is lighter in acidic environment,and a deep brown color in alkaline environment.K+,Na+,Ca2+had no effect on the extract solution;Mg2+,Zn2+had little effect;Fe3+,Cu2+,A13+ had greater effect.The scavenging rate of DPPH free radicals was slightly lower than that of VC,and the total reduction of iron ions was slightly higher than that of VC.
