中文摘要: 以菠萝皮渣粗多糖为原料,对其脱蛋白和脱色工艺进行了探讨,并评价了所得精多糖的抗氧化能力。以脱蛋白率和多糖保留率为评价指标,对盐析法、三氯乙酸(trichloroacetic acid,TCA)法、Sevag法、TCA-正丁醇法、TCASevag法、聚酰胺法和酶法的脱蛋白效果进行对比,筛选最佳的脱蛋白方法;采用活性炭对脱蛋白后的多糖溶液进行脱色,在单因素试验的基础上通过正交试验优化脱色工艺;利用清除DPPH·法和ABTS+法评价所得精多糖的体外抗氧化活性。结果表明:TCA-正丁醇法脱蛋白效果最好,蛋白质脱除率为81.47%,多糖保留率为85.91%;活性炭脱色最佳工艺条件为:活性炭添加量3.0%,脱色温度60℃,脱色时间80 min,样液pH 4.0,此条件下脱色率和多糖保留率分别为64.60%和83.97%。所得精多糖对DPPH·和ABTS+自由基均有一定的清除作用,随着质量浓度的增加,清除率逐渐增大,当浓度为3.0 mg/mL时,清除率分别达到83.27%和49.12%。筛选方法可有效去除菠萝皮渣粗多糖中的蛋白质和色素,所得精多糖具有良好的抗氧化作用。
Abstract:Using crude polysaccharides from pineapple pomace as the raw material,the deproteinization and decoloration process conditions were studied and the antioxidant activity of the purified polysaccharides were also investigated.Seven methods were used to select the optimal deproteinization method with the rate of deproteinization and polysaccharide retention as indexes,including salting out method,trichloroacetic acid(TCA)method,Sevag method,TCA-n-butanol method,TCA-Sevag method,polyamide method and enzymatic method.Then,activated carbon was used to decolorize of polysaccharide solutions after removal of protein,and the conditions was optimized by orthogonal test on the base of single factor experiments.Results indicated that the method of TCA-n-butanol had the ideal deproteinization effect,the deproteinization rate and polysaccharide retention rate were reached to 81.47%and 85.91%,respectively.The optimum decoloration conditions were added 3.0%active carbon at sample pH value of 4.0,with temperature at 60 °C for 80 min,the decolorization rate and polysaccharide retention rate were 64.60%and 83.97%,respectively.The purified polysaccharides possessed scavenging capacity against DPPH·and ABTS+radicals with a dose-dependent relationship.The concentration of 3.0 mg/mL scavenged 83.27%DPPH·and 49.12%ABTS+radicals,respectively.The results demonstrated that these methods can efficiently remove protein and pigment from pineapple pomace polysaccharides,and the purified polysaccharides had stronger antioxidant activities.
