中文摘要: 以茯苓提取粗多糖为原料,考察7种大孔树脂纯化茯苓提取粗多糖的效果。通过静态吸附-洗脱试验结果表明,AB-8型大孔树脂对茯苓提取粗多糖的脱色率与多糖回收率均优于其它种类树脂。通过动态吸附-洗脱试验结果,得到最佳纯化茯苓提取粗多糖的工艺条件为:配制5.0 mg/mL的茯苓粗多糖提取液,以2 BV/h流速上样至柱体积为7 BV的AB-8型大孔树脂内吸附,随后采用5 BV的50%乙醇溶液,以2 BV/h流速洗脱。通过定量分析结果表明,在最佳纯化工艺条件下,茯苓提取粗多糖的脱色率达到85.2%,多糖回收率为75.4%。

Abstract:The seven types of macroporousresins were studied in order that observe effects of decolorizationand retention of polysaccharides in poria cocos extract.Through static adsorption and desorption experiment,AB-8 resin was found to be suitable for the purification of polysaccharides from poria cocos extract.By using dynamic adsorption and desorption experiments,the optimum purification conditions were obtained as follows:5.0 mg/mL poria cocos extract was loaded on the resin column which the volume was 7 BV at flow rate of 2 BV/h and eluted with 5 BV of 50%ethanol at flow rate of 2 BV/h.Under the optimum conditions,the resulting decolorization and polysaccharides retention rates of poria cocos extract was 85.2%and 75.4%,respectively.
