中文摘要: 以阿克苏骏枣为原料,采用生物复合酶解对骏枣多糖进行提取,以多糖得率和纯度为指标,探讨酶解温度、酶 解时间、酶解 pH、加酶量对多糖得率和纯度的影响,通过响应面分析法优化骏枣多糖最佳提取工艺,并探讨骏枣多糖的 聚酰胺柱层析脱色工艺条件。确定骏枣多糖最佳的生物复合酶解提取工艺条件为:酶解温度 60 ℃,酶解时间 65 min,pH 6,加酶量 1 %。在此条件下,多糖得率为 12.29 %,纯度为 39.23 %。聚酰胺柱层析最佳脱色工艺为:聚酰胺用量为骏枣粗 多糖的 4 倍,用 1 倍柱体积的去离子水以 1.5 mL/min 的流速进行洗脱。

Abstract:Akesu Jun jujube was used as raw material, polysaccharides from Aksu Jun jujube were extracted using biological compound enzyme hydrolysis . The yield and purity of polysaccharides were used as index, Ef- fect of enzymolysis temperature, enzymolysis time, pH, enzyme concentration on the yield and purity of polysaccharides was explored.The extraction technology of polysaccharides from Aksu Jun jujube was optimized through response surface analysis.Decoloration of polysaccharides using polyamide column chromatography was also discussed.The optimum extraction was as follows: enzymolysis temperature was 60 ℃, hydrolysis time was 65min, pH was 6, enzyme volume was1 %.Under the optimum extraction technology, The yield and purity of polysaccharides were 12.29 % and 39.23 %,respectively.The optimum decolorization technology was as follows: The content of polyamide was 4 times of crude polysaccharides, and the polyamide column was eluted with 1 column volumes of deionized water at flow rate of 1.5 mL/min.
