中文摘要: 通过单因素试验和正交试验对冬虫夏草产分生孢子培养基进行优化,同时探讨培养条件(温度、光照和 pH)对 分生孢子产量的影响,确定冬虫夏草菌产分生孢子的最佳条件。结果显示:最佳的冬虫夏草产孢子培养基组成为:葡萄 糖 30 g,酵母粉 10 g,CaCl2·2H2O 0.3 g,MgSO4·7H2O 0.5 g,KH2PO4 1.0 g,NaCl 1.0 g,FeSO4·7H2O 0.05 g,ZnCl2 0.09 g,组氨酸 0.3 g,赖氨酸 0.3 g,纯牛奶 200 mL,超纯水补充至 1 000 mL。最佳培养条件为:14 ℃、pH5.80、完全黑暗的环境下,培养 40 d。在最佳培养基和最佳培养条件下,冬虫夏草单菌落分生孢子产量达到 2.98×105 个,优化后孢子产量是基础培养基 孢子产量的 3.6 倍,明显提高了分生孢子的产量。

Abstract:Using the single factor and the orthogonal test, the optimal culture medium for the conidia of Ophiocordyceps sinensis were studied. The effect of temperature,pH and light on the conidia production were also studied. The best culture medium recipe was: glucose 30 g, yeast extract 10 g,CaCl2·2H2O 0.3 g, MgSO4· 7H2O 0.5 g,KH2PO4 1.0 g, NaCl 1.0 g,FeSO4·7H2O 0.05 g, ZnCl2 0.09 g,histidine 0.3 g lysine 0.3 g,pure milk 200 mL, the ultrapure water added to 1 000 mL. The maximum conidia production was produced under the optimal conditions: temperature 14 ℃ , pH 5.80 and complete darkness for 40 d. Under the conditions, the maximum conidia production was 2.98×105. After optimization, the conidia production was 3.6 times more than that of the basal culture medium. The conidia production was significantly increased after optimization, which might be beneficial to workload in scale up fermentation later.
