中文摘要: 建立一种反相高效液相色谱(RP-HPLC)法测定竹茹提取物及其不同部位的腺苷含量的方法。采用RP-HPLC法;Luna C18 ODS柱(4.6?250mm,5 μm),乙腈-0.01 mol/L磷酸缓冲溶液(pH=6.5);体积流量:1.0 mL/min;进样量:10 μL;柱温:40 ℃;检测波长:257 nm。结果 采用RP-HPLC法竹茹提取物及其不同部位的腺苷含量,在0.121–124 μg/mL范围内线性关系良好(R2= 0.9999),平均回收率为98.3%,RSD为3.11%。本方法简便、灵敏、准确,可普遍应用于竹茹提取物的质量研究和品质控制。

Abstract:To develop a method for determination of adenosine in extract of bamboo shavings by RP-HPLC. RP-HPLC method was used with Luna C18 ODS (4.6?250mm, 5 μm) column. The mobile phase was acetonitrile -0.01 mol/L phosphate buffer solution(pH=6.5) with 1.0 mL/min flow rate; The column temperature was 40℃; The detection wavelength was set at 257 nm. RP-HPLC method was used for the determination adenosine; The calibration curve of adenosine was linear under the content 0.12–124 μg/mL; The linearity the average recovery was from 98.3%, RSD was 3.11%. Conclusion The method is simple, sensitive and accurate. The method can be adapted to control the quality of extract of bamboo shavings.
