中文摘要: 为获得具有较高抗冻活性的肽段,以脱脂南极磷虾为研究对象,选择碱性蛋白酶作为最佳蛋白酶,并对其酶解产物的氨基酸组成进行分析。利用超滤、Sephadex G-25凝胶过滤层析、液相色谱-串联质谱联用(liquid chromatography-tandem mass spectrometry,LC-MS/MS)对脱脂南极磷虾抗冻肽(antifreeze peptides from defatted Antarctic krill,AKAPs)进行分离、纯化与鉴定,同时通过测定冻融前后的过氧化氢酶活力评价其抗冻活性的变化,通过测定对鼠李糖乳杆菌的低温保护能力进一步验证合成后AKAPs的抗冻活性。结果表明:AKAPs中富含脯氨酸(20.67%)、丙氨酸(9.53%)和甘氨酸(9.24%)。AKAPs经分离纯化后,含小分子肽较多的组分具有良好的抗冻活性,抗冻活性得到了显著提高(10.12%)。LC-MS/MS鉴定出六肽DINFRY(DY)和九肽LGDGMSLST(LT)。生物合成DY和LT并互溶得到复合肽段DY+LT。冻融后DY、LT、DY+LT处理组鼠李糖乳杆菌的相对存活率显著高于生理盐水处理组(P<0.05),并表现出比商业抗冻剂更好的低温保护作用。表明脱脂南极磷虾中具有较高抗冻活性肽段的氨基酸序列是Asp-Ile-Asn-Phe-Arg-Tyr、Leu-Gly-Asp-Gly-Met-Ser-Leu-Ser-Thr。
Abstract:To obtain peptides with high antifreeze activity,the degreased Antarctic krill was studied with alkaline protease as the optimal protease. The amino acid composition of its hydrolysate was analyzed. Antifreeze peptides from defatted Antarctic krill (AKAPs) were isolated,purified,and identified using ultrafiltration,Sephadex G-25 gel filtration chromatography,and liquid chromatography-tandem mass spectrometry (LC-MS/MS).The changes in the antifreeze activity were determined by measuring the enzyme activity of hydrogen peroxide before and after a freeze-thawing cycle. The antifreeze activity of biosynthesized AKAPs was further verified by determining the cryoprotective ability against Lactobacillus rhamnosus. The results showed that AKAPs were enriched in proline (20.67%),alanine (9.53%),and glycine (9.24%). After isolation and purification of AKAPs,the fraction containing more small molecular peptides had strong antifreeze activity. The antifreeze activity was significantly improved (10.12%). The hexapeptide DINFRY (DY) and the nonapeptide LGDGMSLST (LT) were identified by LC-MS/MS. DY and LT were biosynthesized and intercalated to obtain the composite peptide DY+LT. The DY,LT,and DY+LT treatment groups after freeze-thawing cycle were significantly higher than the saline treatment group(P<0.05) in the relative survival rates of L. rhamnosus,with better cryoprotection than commercial cryoprotectants. It was shown that the amino acid sequences of AKAPs are Asp-Ile-Asn-Phe-Arg-Tyr,Leu-Gly-Asp-Gly-Met-Ser-Leu-Ser-Thr.
