芍药花多酚的纯化及其抗氧化活性研究

中文摘要: 研究大孔吸附树脂纯化芍药花多酚的条件和纯化后多酚的抗氧化能力。采用静态试验和动态试验确定纯化条 件,计算纯化后多酚的含量。在供试的 9 种大孔吸附树脂中,HPD100 树脂的吸附和洗脱效果最好。在室温下,以 50 %乙 醇为洗脱剂, 吸附流速和洗脱流速均为 10 mL/min 时, 纯化的芍药花多酚含量为 721.72 mg GAE/g DW, 是未纯化前的1.57 倍。通过 DPPH、FRAP 和 ABTS 三种方法测得纯化后芍药花多酚的抗氧化能力是未纯化前粗提物的 1.35、1.44、1.62倍。

Abstract:Using the macroporous adsorptive resins, the purification conditions for total phenolic content(TPC) from the flower of Paeonia lactiflora Palla and the antioxidant capacity of the TPC were studied after purification. Static and dynamic experimental methods were performed to determine the purification conditions, and the con- tents of the purified TPC were calculated. Among the 9 macroporous resins tested, HPD100 resin was the best in adsorption effect and desorption effect. At room temperature, when 50 % of ethanol was used as desorption solu- tion, adsorption and desorption flow velocities were both 10 mL/min, the content of the TPC purified from the flower of Paeonia lactiflora Palla was 721.72 mg GAE/g dry weight, and it was 1.57 times higher compared with that before purification. The antioxidant activities of purified extracts measured by 2,2-diphenyl-1-picrylhy- drazyl radical scavenging activity assay(DPPH), ferric reducing antioxidant power assay(FRAP), and ABTS+ radical scavenging activity (ABTS)assay were 1.35, 1.44 ,1.62 times higher than those of crude extracts before purification, respectively.
