松毛菇多酚的提取及纯化工艺研究

中文摘要: 以松毛菇为原料,研究液料比、提取时间、提取温度、超声功率、乙醇浓度、提取次数对松毛菇多酚提取效果的影响,在此基础上利用响应面法优化超声波辅助提取松毛菇多酚的工艺条件,并以吸附及解吸效果为评价指标,筛选出最适大孔树脂,确定其纯化松毛菇多酚的最佳工艺。结果表明,松毛菇多酚最佳提取工艺为液料比22 ∶1(mL/g)、提取温度67 ℃、乙醇浓度71%,在此条件下松毛菇多酚提取量为14.60 mg/g。优选D101 树脂为松毛菇多酚纯化的最适大孔树脂,其对松毛菇多酚的最佳纯化工艺为上样液pH 3,洗脱剂乙醇浓度50%,上样流速0.5 mL/min,上样浓度0.8 mg/mL,洗脱流速1 mL/min,洗脱剂用量4 BV,纯化后多酚含量为(71.18±0.9)mg/g,比粗提多酚提高了近3.9 倍,表明纯化效果良好。
Abstract:Using Lyopyllum cinerascens as raw material,the influence of liquid-solid ratio,extraction time,extraction temperature,ultrasonic power,ethanol concentration and extraction times on the extraction effect of polyphenols from Lyopyllum cinerascens was studied.On this basis,the response surface method was utilized to optimize the process conditions of ultrasonic assisted extraction of polyphenols from Lyopyllum cinerascens.With adsorption and desorption effect as evaluation index,the optimum macroporous resin was selected and the optimum purification process for polyphenols from Lyopyllum cinerascens was determined.The results showed that the optimum extraction process was as follows:liquid-solid ratio 22 ∶1(mL/g),extraction temperature 67 ℃and ethanol concentration 71%.Under these conditions,the extraction amount of polyphenol was 14.60 mg/g.The D101 resin was selected as the optimum macroporous resin for the purification of polyphenols from Lyopyllum cinerascens.The optimum purification process of the polyphenols from Lyopyllum cinerascens was as follows:sample pH 3,ethanol concentration 50%,sample flow rate 0.5 mL/min,sample concentration 0.8 mg/mL,elution rate 1 mL/min and elution dosage 4 BV.After purification,the content of the polyphenols was (71.18±0.9)mg/g,which was nearly 3.9 times higher than that of crude polyphenols,indicating that the purification effect was good.
