中文摘要: 为提高植物甾醇酯的应用价值,在传统酶催化合成方法基础上,研究一种新型的催化剂。以纳米管为乳化剂,利用异辛烷和含皱褶假丝酵母脂肪酶溶液构建Pickering 乳液催化剂(CRL@PE),借助光学显微镜、扫描电镜和共聚焦显微镜对该乳液催化剂形貌进行分析,确定其液滴直径为40 μm~60 μm 的油包水构型。将CRL@PE 应用于催化植物甾醇和α-亚麻酸酯化反应合成亚麻酸甾醇酯,采用单因素试验分析含水率、酸醇摩尔比、酶用量、反应温度、反应时间对植物甾醇转化率的影响,得到最优反应条件为含水率4.0 %、酸醇摩尔比2 ∶1(植物甾醇30 μmol)、酶用量12 U、温度30 ℃、反应时间12 h,此时植物甾醇转化率为93.2%。CRL@PE 重复使用10 次后,甾醇转化率为90.06%;使用15 次后,转化率为77.3%,显示了良好的稳定性。此外,CRL@PE 还具有良好的热稳定性,但对温度变化较为敏感。
Abstract:In order to promote the application of phytosterol ester,a novel catalyst was developed on the basis of traditional enzymatic synthesis method.Pickering emulsion catalyst (CRL@PE)with two-phase structure was constructed using isoctane and Candida rugosa lipase solution and employing nanotube as emulsifier.CRL@PE was identified as oil/water type with the droplet diameter ranging from 40 μm to 60 μm by optical microscopy,scanning electron microscopy and confocal microscopy.Then CRL@PE was used to catalyze the synthesis of βsitosterol linolenic acid ester with β-sitosterol and α-linolenic acid as substrates.The effects of water fraction,molar ratio of phytosterol to linolenic acid,enzyme dosage,temperature and reaction time on the conversion efficiency of phytosterols were studied by single factor experiment.The maximum conversion efficiency was 93.2 % under the optimal process parameters:water fraction of 4.0 %,molar ratio of linolenic acid to phytosterol of 2 ∶1(phytosterols of 30 μmol),enzyme dosage of 12 U,temperature of 30 ℃,and reaction time of 12 h.The conversion efficiency of phytosterol was 90.06% after CRL@PE reused 10 times and 77.3 % after 15 times reused,showing CRL@PE owned good stability.In addition,CRL@PE also had good thermal stability,but was more sensitive to temperature changes.
